Nanosensor Based Determination of Total Cholesterol in Food Samples

Cholesterol is a structural component of biological membranes. It is present in egg yolk, dairy products, goat meat, beef meat, chicken meat etc. Cholesterol is an essential biomarker in the clinical diagnosis of hypertension and arteriosclerosis, which can initiate into other clinical disorders, such as coronary heart disease, myocardial infarction, cerebral thrombosis and even Alzheimer’s disease. A higher level of cholesterol is linked to diabetes mellitus, nephrosis and hypothyroidism, etc., while a lower level of cholesterol is observed as a diagnostic parameter of malabsorption, anaemia, hyperthyroidism, Alzheimer’s disease, etc. The traditional method for determination of cholesterol is found very poor specificity, instability of color forming agent and standardization difficulties.


We can be developed a new kind of matrix for the immobilization of Cholesterol oxidase (ChOx) toward fabrication of a cholesterol biosensor. The new matrix consists of multi-components such as (i) gold (Au) particles distributed MWCNT and (ii) Chitosan (Chi) (a biopolymer) (iii) and an ionic liquid (IL). On the fabrication of a cholesterol biosensor by combining the advantages features of MWCNT, Au nanoparticles, Ch and IL are used. A biosensor electrode was fabricated by entrapment of ChOx onto MWCNT(SH)–Au/Chi–IL matrix and the biosensor is designated as MWCNT(SH)–Au/Chi–IL/ChOx. The existence of Chi–IL adds stability and reproducibility to the biosensor. The MWCNT(SH)–Au/Chi–IL/ChOx biosensor can be exhibited an excellent linear response to cholesterol (Ch) for a wide concentration range with good selectivity and sensitivity. The combinations of MWCNT–metal particle composite along with Chi-IL composite will be developed as the matrix for the construction of cholesterol to apply for the detection of total cholesterol in meat samples. The developed cholesterol nanosensor can be applied for monitoring food quality control as consumer protection and also for forecast awareness in the society before buying from the local market.
